Arriaga Department of Chemistry, California State polytechnic University, Pomona, CA, 91768 USA This work reports on a new individual organelle analysis procedure to i ; sample mitochondria from a tissue cross section into a fused silica capillary, ii ; on-column label these organelles with a mitochondrion-selective probe, and iii ; analyze the labeled organelles by capillary electrophoresis with post-column laser-Induced fluorescence detection CE-LIF ; . In this procedure the bright field visualization and positioning of the sampling end of the capillary is not compromised by the fluorescence of a labeling reagent in the tissue cross section. In addition, oncolumn labeling allows for better control of the reaction time and probe concentrations. As a proof-of-principle, we show results of mitochondria from rat gastrocnemius muscle cross sections that were on-column labeled with 10-N-nonyl acridine orange, a mitochondrion-specific probe that stoichiometrically binds to cardiolipin. The CE-LIF results i.e., organelle number, electrophoretic mobility of individual organellar events, and relative cardiolipin content of individual organellar events ; for the gastrocnemius cross sections are similar when labeling is performed either oncolumn or in-tissue, further confirming the utility of the procedure. On-column labeling of organelles not only makes more practical the CE-LIF analysis of muscle mitochondria in aging and exercise physiology studies, but may easily be extended to the analysis of individual organelles in other biological samples.

Former Director of the Imperial Cancer Research fund neatly summarized this in regard to cancer: "despite the genetic predisposition, the tumor requires an environmental [trigger] as we have known for 200 years that chemical abuse leads to cancer."92 Hence just because there may be more people carrying a genetic susceptibility to ALS, it does not mean we have to ignore exogenous factors. We agree with Riggs & Schochet that there has been a major reduction in ischemic heart disease and stroke, but disagree that "there is no need to invoke intrinsic etiological alterations in the environment, " as comforting such a position might be for some people.93 There are a number of flaws in Riggs position. For example Riggs used the Gompertzian analysis to account for the rising cancer mortality in the USA between 196287 and argued that this was "evidence against environmental causation." Yet he virtually ignored the fact that since the 1960's, quite apart from the major improvements in the treatment of cancers and the extended fiveyear survival rates, the incidence of malignancy in younger adults, under 30 has been rising in most Western countries.94 This trend continued until we saw the impact of reduced smoking positively influencing the prevalence rates.95 Nonetheless, if you attend a chemotherapy center you will find that one in seven patients is less than 35 years old, which was unprecedented 30 or more years ago. We may being unfair to the Gompertzian approach, but some use it as a defeatist position in regard to ALS and the other degenerative diseases, and it is an oversimplification of the surely redundant argument of `nature versus nurture.'96 Moreover, the time period that Riggs examined ALS data is, in genetic terms, quite a short period and the Gompertzians make no attempt to account for this apparent speeding up of the genetic influence in the national gene.
